![]() ![]() The results of this study demonstrate that CGB scaffolds are biocompatible with hUC-MSCs and that granular CGB scaffolds covered with hUC-MSCs are expected to generate new advances for future treatment of traumatic brain injury.Ĭrown Copyright © 2012. BDNF released from CGB scaffold promoted neuronal differentiation of NSCs and led to significant differences in differentiation rate and average neuron perimeter compared with the control group. In this review, we provide a comprehensive overview of CS structure and its physicochemical and biological properties, which have been employed as printing materials to fabricate 3D scaffolds. Chitosan is especially attractive as a bone scaffold material because it supports the attachment and proliferation of osteoblast cells as well as formation of. Chitosan (CS) has been used extensively as a biopolymer for biomaterials used in bone tissue renewal 39, 40. The CGB scaffolds released BDNF in a uniform profile, whereas the CB scaffolds only released BDNF during the first 3 days. Chitosan, a biodegradable, naturally occurring polymer, has drawn considerable attention in recent years as scaffolding material in tissue engineering and regenerative medicine. There were on average more cells on the CGB scaffold on the first day than on any other day sampled (P < 0.05). There were no significant differences in diameter and porosity of individual CGB scaffolds (P > 0.05). Finally, we investigated whether the released BDNF can induce NSC to differentiate into neurons. The amount of BDNF released from CGB over a 30 day period was determined by ELISA. Prior to this, the diameter and porosity of CGB scaffolds were measured. The viability of hUC-MSCs adhered to the CGB scaffold was determined by digesting with 0.25% trypsin and evaluating with the cell counting kit-8 (CCK-8). Fluorescent nuclear staining (Hoechst 33342) was employed to determine the attachment of the hUC-MSCs to CGB scaffolds on the 1st, 3rd, 7th and 10th day of co-culture. hUC-MSCs were co-cultured with the CGB scaffold. The collagen/chitosan scaffold was more effective to promote and accelerate L929 cell proliferation, particularly for scaffolds containing 30 of chitosan. The chitosan scaffold based on immobilization of BDNF by genipin (GP) as a crosslinking agent referred to hereafter as a CGB scaffold was prepared by freezing-drying technique. This study tested the cytotoxicity of a BDNF blended chitosan scaffold with human umbilical cord mesenchymal stem cells (hUC-MSCs), and the in vitro effect of BDNF blended chitosan scaffolds on neural stem cell differentiation with the aim of contributing alternative methods in tissue engineering for the treatment of traumatic brain injury (TBI). ![]() The Adobe reader supports viewing, printing and annotating of PDF files while the Acrobat is used to create, edit, convert, digital sign, encrypt and export PDF files. This was a more enjoyable setup than the first two not as harsh in the treble and the stage was a little further back. Setup 3: Element X -> Echo -> Phonitor -> Utopia I didn’t spend much more time with this setup. Next I decided to try a funk track (“Doo Wa Ditty”)… this was a mistake with this setup, the “talk-box” sound and harmonica (i think) were painfully sharp. “Song of the Caged Bird” by Lindsey Stirling sounded solid, airy, and not harsh. It was more dynamic than “Setup 1”, punchier but… Too harsh in the treble for much listening for me. Vocals more centered, stage good and strong. I came in thinking this setup would be good, borderline phenomenal… I was disappointed. Setup 2: Element X -> Feliks Echo -> Utopia A clean listen but not necessarily “enjoyable” Leans bright, maybe too bright, depending on the track some sounds (claps for example) came in kinda hot. Very clear/transparent, good stage (width/depth). Setup 1: Matrix Element X -> SPL Phonitor -> Utopia Build quality is typical Focal build which looks and feels strong and high quality. I suspect it’s done to create a luxury experience and, well, it does feel like you bought something significant unboxing it. Look & Feel: The Focal presentation always seems to start with what I’ll call somewhat oversized but nicely done packaging. The sound was often brighter than I like and treble would get in the range of being rather piercing and fatiguing. The problem I had was getting to that “best” as I didn’t think it paired well with many of my amps (to my liking, at least). General thoughts / The TL DR: In my experience, at its best, the Utopia provided a clean, strong, and unrestrained view into my music. I really appreciate the opportunity to try out a flagship headphone like the Utopia and you guys engagement with the community. That experience has lead me to never wanna buy a McIntosh product ever.įound my write up… Focal Utopia Impressionsįirst off, thanks to Taron, Andrew, and Tyler for providing the Utopia and coordinating getting them around to all the people to be demoed. I am serious, I have a $50 dollar ChiFi AIO that has better quality knobs than the MHA150 both in actual material (MHA150 is the cheapest thinnest plastic you will ever feel) as well as actual pot (MHA150 is the loosest pot I have ever felt, you aren’t careful and you turn way too much and you blew out your eardrums and exploded your headphones). ![]() Let me tell you, the MHA150 has the cheapest knobs I have ever felt. The thing that would make me never buy a McIntosh product is their knobs. People are buying these because they have money, want a system, and don’t know better. People see them and think high quality and high end, but really it was all built from their past, with most of their current products being pretty trash. They are getting by because they are a “luxury” brand for HiFi now. Since they kind of went full commercial, the new build quality is sub par on most of their products, they sound not so good, and are just way overpriced for what they are. People need to see that Utopia is real.I have heard and felt some modern McIntosh vs vintage McIntosh and I would say they are working on past glory. They can create energy that spews out magical things - new cures, new buildings, new avenues for people to move forward. ![]() With every album I live in these worlds in my mind - I’m trying to show people experiences where utopian things can exist, and you can enjoy yourself and have a good time. The final thing I will be doing is enabling the native MAC Learning functionality that is now built into vSphere 6.7 so that I do not need to enable Promiscuous Mode, which has normally been a requirement for the Nested portgroup and nested labs in general. I’ll now also be able to access the ESXi UI and VCSA UI, once they are stood up. I’ve configured a route summary of 10.254.0.0/16 to go through the IP address of the WAN interface as the gateway so that I can access the Admin UI at as well. This is where the static route will now be useful to access your nested lab. Choose something easy to label them as which makes sense to you: Navigate to Firewall > Add firewall rule and create the following rules. With our VLANs created, we’ll need to create two firewall rules to allow traffic from the WAN port to access the LAN, as well as to allow traffic from LAN to LAN. Navigate to Networking and select Add Interface > VLAN to create each of your networks. Now, we can create our VLANs for our nested environment. Once in the Admin GUI, navigate to Administration > Device Access and tick the box for WAN under the HTTPS column. ![]() The WAN port is set to grab an address from DHCP so you’ll need to determine which IP address this is either by going into your physical router, or using a tool like Angry IP. To do so, choose option 4 to enter the device console and enter the following command: system appliance_access enable So what do we do? We need to run a command to enable admin access on the WAN port. Unfortunately, this will not work since the LAN side has no physical uplinks. Once this is done, you would normally navigate to that address on port 4444 to access the admin GUI. Login to the console with the default ( admin – admin) credentials, and choose the option for Network Configuration to change the IP for your nested LAN port. This will need to be changed to the subnet you’re using for your nested LAN interface. Once the VM has been deployed, the Sophos XG will be configured with a 172.16.1.1 address by default. I have a bunch of VLANs created for my physical Home Lab as I’ve yet to deploy NSX-T in there, but once I do, I’ll be removing the majority of said VLANs and only keeping the required ones needed to run the lab.
This 2022 report states that IT organizations still have challenges to solve to break knowledge silos and improve their inner source strategy. “Over 48% of respondents regardless of function, think onboarding takes a very long time or a somewhat long time at their organization.” The results set contains over 80,000 responses from almost every country on Earth. This process can take up to several months: Knowledge access is crucial during onboarding. But indirectly, knowledge silos generate a loss of productivity. That’s part of developers’ work to help and interact with each other. “46% of all respondents spend more than 30 minutes a day answering questions.” So when we don’t find answers, we talk to people: “About a quarter of all respondents say they can’t find up-to-date information within their organization to help them do their job and they aren’t able to quickly find answers to their questions with existing tools and resources.” Knowledge should be more accessible, but it should be relevant. “Only 16% of organizations have Innersource initiatives.” That’s not surprising if we consider that: ![]() “68% of respondents say they encounter a knowledge silo at least once a week.” So there are challenges to improving knowledge transfer and code/ practices reuse in the IT companies. And last but not least Bradley Whitford was wonderful in his short performance. I also liked Judith Light whose role was a bit cartoonish but fun. Her singing number is the peak of the film but also the performance around it is filled with a great amount of naturalism and convincing from the first to the last minute she is on screen. Alexandra Shipp rounds the cast up with a wonderful turn. Glad he is finally getting the attention he deserves. He already made my Top 5 last year with his tremendously underrated performance in "The Boys in the Band". Robin de Jesus once again pulled out a fantastic supporting performance. With the charm and looks of a young Tom Hanks, Andrew Garfield really and once for all pushed himself to the firmament of Hollywood. Not only can he sing brilliantly but the way he embodied that role, captured and expressed the emotions and really developed into it was just out of this world and can easily marked down as one of the best performances of the year. Its contains a sensational performance by Andrew Garfield in the role of Jonathan Larson. An emotional musical about ambition, friendship and just living towards your dreams. A great portrayal of an artist that left the stage far too soon. And finally a movie that really went under my skin this year. It is his career best performance to date. He is frenetic, loud, theatrical, authentic, funny, heartbreaking and authentic. But above all Garfield is *perfect* in this. I don't like how a straight guy is again at the center of an AIDS story. But you can lob some of the same criticism at this as you can RENT-the material has a blinkered idea of what selling out is, there's some questionable characterizations. ![]() People who generally don't like musicals might find this one's scale and intimacy appealing. ![]() The intercutting of the story, show and home movies reminded me a little of Chicago (2002) or All That Jazz (from the 70's) in how the songs are woven around the plot. The formal editing and direction choices-and how the script motivates the various vignettes-makes the film feel alive and the songs makes sense in universe. The film does a *really* good job of turning what began life as a monologue into a cinematic story. The music and general plot outline is Larson's most engaging work. So this adaptation starts from a better place than the RENT film a few years back. Tick, Tick.Boom is a more vivid and authentic work. I find the show's treatment of AIDS especially distasteful. I think RENT (in any form) is a pretty poor musical, regardless of how you look at it. It’s a great feature that users always look forward to using with the Flashpaq F5.
